Trityl Resins

Trityl resins are the most versatile resins for solid phase synthesis. These supports are used for the immobilisation of carboxylic acid, alcohol, phenol, amine, thiol, and imidozole. Almost any nucleophilic function can be linked to these resins under extremely mild conditions [1, 2]. Furthermore, cleavage of the products from these resins takes place also under very mild conditions. In this trityl resin family, 2-chlorotrityl resin is the least acid labile, and 4-methoxytrityl resin is the most acid labile.

Attachment of carboxylic acid on trityl chloride resin [1]: In a typical experiment, carboxylic acid (6 mmole) and DIEA (4.4 ml, 25 mmole) are dissolved in dry dichloromethane (100 ml). A small amount of DMF is added if the acid is not completely dissolved in DCM. The resulting solution is added to trityl chloride resin (10 g). The suspension is stirred for other 20-120 minutes. Subsequently methanol (10 ml) is added, and stirring is continued for other 10 minutes. The resin is filtered, washed (with 3 x DCM, 2 x DMF, 2 x methanol), and dried in vacuum over KOH.

Attachment of alcohol and phenol on trityl chloride resin [1]: The alcohol or phenol (2 eq.) and pyridine [4 eq.] dissolved in anhydrous THF (10 ml per gram of resin) are added to trityl chloride resin. The suspension is stirred at 60 0C for 2 - 6 h. At end of the reaction, the resin is filtered, washed with 3 x DCM/MeOH/DIEA (17:2:1), 3 x DCM, 3 x DMF, 3 x methanol, and dried in vacuum over KOH.

Attachment of amine on trityl chloride resin [1]: The amine (2 - 4 eq.) dissolved in anhydrous THF (10 ml per gram of resin) is added to trityl chloride resin. The suspension is stirred for 2 - 6 h. At end of the reaction, the resin is filtered, washed with 3 x DCM/MeOH/DIEA (17:2:1), 3 x DCM, 3 x DMF, 3 x methanol, and dried in vacuum over KOH.
