Fmoc-Amino Acids Attached to Wang Resins

Fmoc-amino acid Wang resins are used for the Fmoc solid phase synthesis of peptide acids. The resins consist of 1% cross-linked polystyrene beads which have been functionized with the TFA labile p-benzyloxybenzyl alcohol linker. All following Fmoc-amino acid Wang resins are prepared by the method of Sieber [1], which gives less enatiomerization and dipeptide formation than those generated by using conventional DCC/DMAP method. The substitution rate of amino acid on resin is determined by photometric measuring the Fmoc-chromophore liberated upon treatment with piperidine/DMF.
